Сімнадцятирічний Кенджі Койсо – юний програміст і математик – почувається в ОЗ набагато впевненіше, ніж в реальності. І під час літніх канікул пише код для одного з віртуальних сервісів мережі. Одного дня Нацукі-семпай пропонує Кейджі альтернативний підробіток – поїхати з нею до маэтка Джіноучі, який розташований у префектурі Наґано, на святкування 90-річчя матріарха клану з метою допомогти їй у домашніх справах. Spinning off from the popular Professor Layton puzzle games, this movie brings Layton and his trusty assistant Luke to the big screen to allow fans to enjoy more of the same. At the beginning of the film Layton receives a letter from the famed opera singer, Jenice Kaitly, inviting them to see her newest performance. She also asks Layton, Luke, and Remi for assistance in a mystery, saying her friend Melina went missing one year ago but returned later as a seven year old girl saying she had found the secret of eternal life. Naturally, the case fascinates Layton and his apprentices, so they head to the Crown Petone opera house. When they arrive, something is very strange: the pianist, Ozro Vislar talks of a game, and the winner will win the "eternal life." It is up to Layton, Luke, and Remi to solve the mystery of the Eternal Diva before it is too late.

He also has to look after his toddler nephew and French-speaking Caucasian sister-in-law, Eloise (Elodie Bouchez), and is fearful that his brother may be tortured in Syria. One night he befriends a passenger, who identifies herself as Phyllis (Robin Wright Penn), claims she is Head of Programming of Q-Dog Television, sympathizes and offers to assist him get his brother released. He gets a shock when she asks him to carry out an act of terrorism as retribution against America. He walks away, and subsequently finds out that she has stolen his money. Shortly thereafter, his cab is confiscated by secret agents, and Eloise is held for questioning. Vengeful, he shaves off his beard, assumes the name of Habib and attends the offices of Q-Dog Television in order to confront Phyllis - not realizing the shock and trauma that await him when he finds out who Phyllis really is.
