Народився у родині будівельника, де крім нього було ще п'ятеро дітей. Сім'я була бідною і неблагополучною, хлопчик кинув школу, крутився у кри ... мінальному середовищі, перебивався випадковими заробітками. Депардьє мав міцну і огрядну статуру, виглядав старшим за свій вік, відповідно і друзів він підбирав собі старших. Від неробства вони спочатку просто хуліганили, брали участь у бійках, покурювали травичку, а потім зайнялися крадіжками та викраденням автомобілів. У Шатору тоді розташовувалася натівська військова база. І місцеві хлопці не раз брали участь у бійках з американськими солдатами. Після однієї з них Жерар Депардьє потрапив у коматозному стані в клініку. Але все обійшлося. Ось тоді Жерар усвідомив, що потрібно щось змінювати у своєму житті, інакше все може закінчитися вельми сумно.
Нічого не сказавши ні родині, ні друзям, Жерар поїхав на південь – до Канн, де влаштувався працювати на пляж. Тут він спостерігав за акторами, які з'їхалися на престижний канський фестиваль. Повернувшись у Шатору, Жерар Депардьє зустрів на вокзалі знайомого, який запропонував йому поїхати в Париж і спробувати себе як актора. У 1965 він приїхав до Парижа і вступив до театральної школи Ш. Дюлена при Національному народному театрі. Там він зустрічає своє перше кохання – однокурсницю Елізабет Гвінот, яка була на 6 років старша за нього (через кілька років, у 1970 вони одружаться). Потім Жерар закінчує Школу драматичного мистецтва Ж.-Л. Коші та разом зі своїми однокласниками Міу-Міу і Патріком Деваєром вступає у трупу маленького аматорського театру «Кафе де ля Гар» («Cafe de la Gare»).
Жерар Депардьє пробував свої сили як режисер на французькій класиці, поставивши «Тартюфа» (1984). Крім того, актор випустив диск з піснями у своєму виконанні, а вина з його виноградників відомі у всьому світі. У 1996 році Депардьє був відзначений орденом Почесного легіону.
